DNA Replication and Polymerase Function

Medium Biochemistry Nucleic Acids And Dna Replication

During DNA replication, various enzymes play crucial roles in ensuring the fidelity and efficiency of the process. One of the key enzymes involved is DNA polymerase, which synthesizes new strands of DNA complementary to the existing strands. However, DNA polymerase cannot initiate the synthesis of a new DNA strand on its own; it requires an RNA primer to start the process. This primer is synthesized by another enzyme called primase.

The ability of DNA polymerase to proofread newly synthesized DNA is vital for maintaining genetic stability. Proofreading occurs in the 3’ to 5’ direction and allows DNA polymerase to correct mismatched bases during replication. If an incorrect base is incorporated, the enzyme can excise it and replace it with the correct base before continuing elongation.
